Tirzepatide

Description

Tirzepatide is a novel dual glucose-dependent insulinotropic polypeptide (GIP) and GLP-1 receptor agonist. It is a 39-amino acid synthetic peptide with a C20 fatty diacid moiety attached for albumin binding, resulting in a long half-life of approximately 5 days. It is approved under the brand name Mounjaro for Type 2 Diabetes.

Mechanism of Action

Tirzepatide acts as a dual agonist on both GIP and GLP-1 receptors. It enhances glucose-dependent insulin secretion, suppresses glucagon secretion, slows gastric emptying, and reduces appetite through central nervous system mechanisms. The dual action provides superior glycaemic control and weight loss compared to single-agonist therapies.
